NEDD8(Neural precursor cell expressed developmentally downregulated protein 8)is an ubiquitin-like protein that is covalently attached to a lysine residue of a protein substrate through a process known as neddylation,catalyzed by the enzyme cascade,namely NEDD8 activating enzyme(E1),NEDD8 conjugating enzyme(E2.,and NEDD8 ligase(E3).The substrates of neddylation are categorized into cullins and non-cullin *** of cullins activates CRLs(cullin RING ligases),the largest family of E3 ligases,whereas neddylation of non-cullin substrates alters their stability and activity,as well as subcellular ***,the neddylation pathway and/or many neddylation substrates are abnormally activated or over-expressed in various human diseases,such as metabolic disorders,liver dysfunction,neurodegenerative disorders,and cancers,among ***,targeting neddylation becomes an attractive strategy for the treatment of these *** this review,we first provide a general introduction on the neddylation cascade,its biochemical process and regulation,and the crystal structures of neddylation enzymes in complex with cullin substrates;then discuss how neddylation governs various key biological processes via the modification of cullins and non-cullin *** further review the literature data on dysregulated neddylation in several human diseases,particularly cancer,followed by an outline of current efforts in the discovery of small molecule inhibitors of neddylation as a promising therapeutic ***,few perspectives were proposed for extensive future investigations.

Metal-organic framework hybrid materials of ZIF-8/RGO for immobilization of D-amino acid dehydrogenase

Immobilization of D-amino acid dehydrogenase(DAADH)by the assembly of peptide linker was studied for the biosynthesis of *** material of zeolitic imidazolate framework-8(ZIF-8)combined with reduced graphene oxide(RGO)was applied for the immobilization of DAADH from Ureibacillus *** recovery rate of DAADH/ZIF-8/RGO was 165.6%.DAADH/ZIF-8/RGO remained 53.4%of its initial activity at 50°C for 10 h while the free enzyme was ***/ZIF-8/RGO maintained 70.5%activity in hyperalkaline solution with pH *** parameters indicated that DAADH/ZIF-8/RGO had greater affinity of phenylpyruvate as V_(max)/K_(m)of DAADH/ZIF-8/RGO was 1.2.-fold than free *** seven recycles,the activity of DAADH/ZIF-8/RGO remained 64.3%.Furthermore,one-step separation and in situ immobilization of DAADH by ZIF-8/RGO/Ni was carried out with 1.5-fold activity *** peptide linker and metal-organic framework(MOF)immobilization,thermostability and activity of the immobilized DAADH were significantly improved.

Low thermal conductivity,compatible thermal expansion coefficient,and good calcium–magnesium–aluminosilicate(CMAS)corrosion resistance are critical requirements for environmental barrier coatings used on silicon-based ***2.i2.7(RE=rare earth)has been widely recognized as one of the most promising candidates for environmental barrier coatings due to its good water vapor corrosion ***,the relatively high thermal conductivity and poor resistance to CMAS corrosion have limited its practical *** by the high entropy effect,in this work,a novel rare earth disilicate(Lu_(_(1/7))Yb_(_(1/7))Sc_(_(1/7))Er_(_(1/7))Y_(_(1/7))Ho_(_(1/7))Dy_(_(1/7)))2.i2.7((7RE_(_(1/7)))2.i2.7)has been designed and synthesized by a solid reaction process.(7RE_(_(1/7)))2.i2.7 showed a low thermal conductivity of 1.81 W·m^(−1)·K^(−1)at 12.3 ***,the thermal expansion coefficient of(7RE_(_(1/7)))_(2.Si_(2.O_(7)(4.07×10^(−6)℃^(−1)from room temperature(RT)to 1400℃)is close to that of the SiC-based ceramic matrix composites(SiC-CMCs)((4.5–5.5)×10^(−6)℃^(−1)).Additionally,(7RE_(_(1/7)))2.i2.7 exhibited excellent resistance to CMAS *** exposed to CMAS at 1300℃for 48 h,the reaction layer thickness was 2.μ*** improved performance of(7RE_(_(1/7)))2.i2.7 highlights its potential as a promising candidate for thermal/environmental barrier coatings.

Quantitative Evaluation of Heavy Metal Pollution and Its Influencing Factors in Water Bodies of Karst Areas

At present, there is relatively little research on the synergistic effects of heavy metals in soil, sediment, and bedrock on heavy metal pollution in water bodies. In this paper, heavy metals in soil, sediment, bedrock, and water of a typical karst watershed in southwest China were investigated. The results indicated that the average values of heavy metals in soil and sediment were relatively higher than those in bedrock except for Ni and As. During the research period, As and Cd were the main heavy metal elements polluting the soil and sediment in the study area, while water bodies were mainly polluted by Ni, As, and Cd. The pollution assessment indicated that there were instances of poor or very poor water quality in the study area during the study period;the soil as a whole was slightly polluted by Cd and As;sediment was subject to moderate Cd pollution and mild As pollution. Analysis of geochemical form for heavy metals showed that heavy metals in soil and sediment were mainly in residual form, and the proportions of exchangeable As and Cd were relatively high. Multiple statistical analysis showed that heavy metals in sediment, soil, and bedrock explained 2..8%, 16.8%, and 16.2. of the changes in heavy metals of water, respectively. The research results can provide scientific basis for the prevention and control of heavy metal pollution in water bodies.

With the rapid development of urbanization,landscape architecture has become an indispensable component of daily *** order to achieve better landscape effects and protect the ecological environment,landscape ecology theory can be integrated into landscape planning to carry out disciplinary crossing and integration research between landscape ecology and landscape *** this study,based on the basic theories of landscape ecology,the waterfront landscape of Dapo Town was designed,with the aim to achieve ecological balance in planning,showcase biodiversity,and create a harmonious landscape ecological environment which can benefit both nature and *** design combined artificial and natural elements while improving the natural environment,which promoted a harmonious relationship between humans and nature,thus enriching the spatial organization of the city,and playing a role in regulating the urban environment and climate.

The current understanding of the legacy and emerging organic pollutants in the soil of Bangladesh remains *** disposal practices,particularly in e-waste,landfills,and industrial operations,may lead to the pervasive presence of persistent organic pollutants(POPs)and polycyclic aromatic hydrocarbons(PAHs)in the *** study aimed to systematically investigate the concentrations,sources,and risk assessment of halogenated flame retardants(HFRs),polychlorinated biphenyls(PCBs),polychlorinated naphthalenes(PCNs),organochlorine pesticides(OCPs),chlorinated paraffins(CPs),and PAHs in the contaminated soil of Dhaka's dumpsites,e-waste,and industrial areas collected in *** total concentration of investigated pollutants averaged at 4790±4040 ng g^(-1)dw,with the following order:P44CPs(4110±4140 ng g^(-1)dw)>P16PAHs(42.±491 ng g^(-1)dw)>P16HFRs(190±538 ng g^(-1)dw)>P2.9PCBs(43.5±73.5 ng g^(-1)dw)>P50PCNs(17.7±2..0 ng g^(-1)dw)>P2.OCPs(9.41±9.61 ng g^(-1)dw).The e-waste site exhibited the highest total concentration of the target pollutants,reaching 12.700 ng g1 *** were the predominant contributors,accounting for averages of 81.8%,70.5%,and 68.5%to waste landfills,e-waste,and industrial sites,respectively.A comprehensive analysis of 2.9 PCB congeners revealed their primary origin from Aroclor PCBs,with minor contributions from unintentionally produced *** highest incremental lifetime carcinogenic risk(ILCR)occurred in e-waste sites,contributed by dioxin-like PCBs,with a maximum value of 5.9×10^(-5)for adults,exceeding the limit 1.0×10^(-6)set by the *** non-carcinogenic hazard quotients(HQs)were much lower,suggesting no significant risk from the investigated *** findings highlight the importance of proper waste management and regulated e-waste recycling to mitigate potential hazardous risks to the Bangladeshi population.

Ecological Remediation Measures for Non-point Source Pollution Based on Source-Sink Landscape Theory: A Case Study of Huanghou Basin

The growth of society and population has led to a range of water pollution *** these,non-point source pollution assessment and treatment pose a particular challenge due to its formation *** has become a focal point and challenge in water pollution treatment *** study area for this research was the Huanghou basin in Guizhou Province,southwest *** land use type of the basin was analyzed using remote sensing technology,and water quality data was collected by distributing points throughout the basin,based on source-sink landscape *** distribution map of the basin’s source-sink landscape and the results of water quality data accurately and efficiently identified the areas with high risk of non-point source pollution in the western and southwestern residential and agricultural areas of the upper ***,a strategy of“increasing sinks and decreasing sources”was *** strategy was implemented at both macro and micro levels to address non-point source pollution in the basin using ecological remediation *** work to control karst rocky desertification should continue at a macro *** rocky desertification area in the basin should gradually transform into grassland and forested land,while increasing the overall area of the sink *** restoration techniques,such as slope planting,riparian zone vegetation restoration,increasing plant abundance,and restoring aquatic plants,can effectively control non-point source pollution at the micro *** to traditional control methods,this remediation strategy focuses on source and process *** is more effective and does not require large-scale water pollution control projects,which can save a lot of environmental control funds and management ***,it has greater research significance and application value.

Fluorinated [2.rotaxanes as sensitive ^(19)F MRI agents: Threading for higher sensitivity

As a promising imaging technology,the low sensitivity of fluorine-19 magnetic resonance imaging(^(19)F MRI)severely hinders its biomedical ***,we have developed an unprecedented rotaxanebased strategy to improve the sensitivity of^(19)F MRI *** threading the fluorinated macrocycle into2.blade pinwheel[2.rotaxanes,the^(19)F longitudinal relaxation rate R1was dramatically increased,resulting in a significant^(19)F MRI signal intensity enhancement of up to 79%.Through comparative molecular dynamics studies using a series of solution and solid-state^(1)H/^(19)F nuclear magnetic resonance(^(1)H/^(19)F NMR)and molecular dynamics simulations,it was found that the formation of mechanical bonds dramatically restricts the motion of the wheel fluorines and thus increasing the R1for higher^(19)F MRI *** a novel strategy for improving^(19)F MRI sensitivity,this study has established^(19)F NMR/MRI as a valuable technology for monitoring the molecular dynamics of rotaxanes,which may shed new light on high-performance^(19)F MRI agents and molecular devices.
