Dynamic shielding of bacterial outer membrane vesicles for safe and efficient chemo-immunotherapy against tumors

Bacterial outer membrane vesicles(OMVs)are potent immunostimulants of regulating the tumor microenvironment(TME)for immunotherapy,and can be used to deliver ***,the severe systemic inflammatory response triggered by OMVs upon intravenous(i.v.)injection has limited their ***,we developed a safe and effective strategy by conjugating doxorubicin-loaded serum albumin(SA-DOX,AD)onto the surface of OMVs using a matrix metalloproteinase(MMP)-cleavable peptide linker(cL).This approach enabled the dynamic shielding of OMVs to reduce the systemic side effects while simultaneously enhancing the anti-tumor effects through ***,the resulting OMV-cL-AD formulation exhibited significantly enhanced accumulation at the tumor site after ***,facilitated by the SA decoration on the OMVs ***,the shield on the OMV-cL-AD was cleaved by the over-expressed MMP in the TME,leading to the release of both OMVs and *** process provided OMV-induced immunotherapy and DOX-induced chemotherapy,resulting in synergistic tumor *** conclusion,our work demonstrated the potential of OMV-cL-AD as an effective immunochemotherapy strategy that can prolong the survival time of mice without inducing side effects.

In the harsh service environment of high temperature and intense neutron irradiation in water-cooled nuclear reactors,the austenitic stainless steel weld overlay cladding on the inner surface of the reactor pressure vessel suffers from thermal aging and irradiation damage simultaneously,which can induce microstructural evolution and hardening of the *** it is quite difficult to achieve this simul-taneous process out of the pile,two kinds of combined experiments,i.e.,post-irradiation thermal aging and post-aging irradiation were performed on 308 L stainless steel weld metals in this *** interactive effect of thermal aging and proton irradiation on microstructural evolution and hardening ofδ-ferrite in 308 L weld metal was investigated by combining atom probe tomography,transmission elec-tron microscopy and nanoindentation *** results revealed that thermal aging could eliminate the dislocation loops induced by irradiation and affect the phase transition process by accelerating spinodal decomposition and G-phase precipitation,thus enhancing hardening of irradiatedδ-*** the effect of irradiation on the microstructure and hardening of thermally agedδ-ferrite,however,intensive collision cascades can intensify G-phase precipitation and dislocation loop formation but decrease spinodal decomposition,leading to a limited effect on hardening of thermally agedδ-***,the interaction of thermal aging and irradiation can promote G-phase ***,the interaction can causeδ-ferrite hardening,which is mainly influenced by spinodal decomposition,followed by G-phase and dislocation loops,where spinodal decomposition and G-phase cause hardening by inducing strain fields.

This study endeavors to formulate a comprehensive methodology for establishing a Geological Knowledge Base(GKB) tailored to fracture-cavity reservoir outcrops within the North Tarim Basin. The acquisition of quantitative geological parameters was accomplished through diverse means such as outcrop observations, thin section studies, unmanned aerial vehicle scanning, and high-resolution cameras. Subsequently, a three-dimensional digital outcrop model was generated, and the parameters were standardized. An assessment of traditional geological knowledge was conducted to delineate the knowledge framework, content, and system of the GKB. The basic parameter knowledge was extracted using multiscale fine characterization techniques, including core statistics, field observations, and microscopic thin section analysis. Key mechanism knowledge was identified by integrating trace elements from filling, isotope geochemical tests, and water-rock simulation experiments. Significant representational knowledge was then extracted by employing various methods such as multiple linear regression, neural network technology, and discriminant classification. Subsequently, an analogy study was performed on the karst fracture-cavity system(KFCS) in both outcrop and underground reservoir settings. The results underscored several key findings:(1. Utilization of a diverse range of techniques,including outcrop observations, core statistics, unmanned aerial vehicle scanning, high-resolution cameras, thin section analysis, and electron scanning imaging, enabled the acquisition and standardization of data. This facilitated effective management and integration of geological parameter data from multiple sources and scales.(2) The GKB for fracture-cavity reservoir outcrops, encompassing basic parameter knowledge, key mechanism knowledge, and significant representational knowledge, provides robust data support and systematic geological insights for the intricate and in-depth examination of the genetic

The Li metal battery with ultrahigh-nickel cathode(LiNi_(x)M_(1.x)O_(2),M=Mn,Co,and x≥0.9)under high-voltage is regarded as one of the most promising approaches to fulfill the ambitious target of 400 Wh/***,the practical application is impeded by the instability of electrode/electrolyte interface and Ni-rich cathode *** we proposed an electron-defect electrolyte additive trimethyl borate(TMB)which is paired with the commercial carbonate electrolyte to construct highly conductive fluorine-and boron-rich cathode electrolyte interface(CEI)on LiNi_(0.9)Co_(0.05)Mn_(0.05)O_(2)(NCM90)surface and solid electrolyte interphase(SEI)on lithium metal *** modified CEI effectively mitigates the structural transformation from layered to disordered rock-salt phase,and consequently alleviate the dissolution of transition metal ions(TMs)and its“cross-talk”effect,while the enhanced SEI enables stable lithium plating/striping and thus demonstrated good compatibility between electrolyte and lithium metal *** a result,the common electrolyte with 1.wt%TMB enables 4.7 V NCM90/Li cell cycle stably over 1.0 cycles with 70%capacity *** work highlights the significance of the electron-defect boron compounds for designing desirable interfacial chemistries to achieve high performance NCM90/Li battery under high voltage operation.

Comprehensive Summary,Regulating spin crossover(SCO)behavior,especially controlling the spin transition steps,is an important scientific issue,mainly because people aim to control spin bistability and ***,SCO bistability can be regulated by changing the ligand-modifying species,non-coordinated anions,guest molecules,and metal-ion ***,the control of multistability is extremely challenging,especially in Fe(III)SCO ***,we report that[FeIII(H-5-Br-thsa)(5-Br-thsa)]·H2O(5-Br-thsa=(5-bromo-2-hydroxybenzylidene)hydrazinecarbothioamide),a compound we have reported before,exists in two polymorphic forms:polymorph 1.exhibits three-step SCO,and polymorph 2 shows five-step SCO,with multi-step SCO behavior effectively regulated by *** to single-crystal and powder X-ray diffractometry,polymorphs 1.and 2 crystallize in different space groups during their spin transitions,with two-step symmetry breaking observed(Pbcn→Pnc2→Pbcn for polymorph 1.P21.n→Pn→P21.n for polymorph 2).We realized that the behavior of these two polymorphs depends significantly on the structure,including(i)the average Fe—N bond distance,(ii)deformation of octahedral FeIII atoms,and(iii)distinct crystal packing,which account for the large differences observed in magnetic properties.

Senile plaque blue autofluorescence was discovered around 40 years ago,however,its impact on Alzheimer’s disease(AD)pathology has not been fully *** analyzed senile plaques with immunohistochemistry and fluorescence imaging on AD brain sections and also Aβ aggregation in *** DAPI or Hoechst staining,the nuclear blue fluorescence could only be correctly assigned after subtracting the blue plaque *** flower-like structures wrapping dense-core blue fluorescence formed by cathepsin D staining could not be considered central-nucleated neurons with defective lysosomes since there was no nuclear staining in the plaque core when the blue autofluorescence was *** Aβ self-oligomers and Aβ/hemoglobin heterocomplexes generated blue *** Aβ amyloid blue autofluorescence not only labels senile plaques but also illustrates red cell aggregation,hemolysis,cerebral amyloid angiopathy,vascular plaques,vascular adhesions,and *** summary,we conclude that Aβ-aggregation-generated blue autofluorescence is an excellent multi-amyloidosis marker in Alzheimer’s disease.
