MnO_(2)-coated nanodiamond-driven photodynamic therapy for enhanced antitumor effect by addressing hypoxia and glutathione depletion

The generation of reactive oxygen species(ROS)at the tumor site to induce destruction is emerging as a novel strategy for cancer treatment,which involves photodynamic therapy(PDT).Nevertheless,tumors typically create a hypoxic environment and are equipped with an endogenous antioxidant defense system that could potentially impede the efficiency of the therapeutic *** overcome these drawbacks,herein,a tumor microenvironment-responsive the ND-PAA-CD-Ce6@MnO_(2)(NPCC@M)delivery system was fabricated by disulfide bond coupling chlorin e6(Ce6)to nanodiamond(ND)and further wrapped by MnO_(2)nanosheets to facilitate *** use of disulfide bond not only stabilizes Ce6 in the blood circulation to prevent premature leakage,but also destroys the antioxidant barrier of overexpressed glutathione(GSH)in tumor ***,the outer MnO_(2)was rapidly degraded by the endogenous hydrogen peroxide(H_(2)O_(2))in the acidic pH and GSH within the tumor cells,which leads to an abundance of O_(2)and while increases the level of 1._(2)under laser *** results eventually broke the redox homeostasis and attenuate hypoxia,thereby inducing apoptosis and necrosis of tumor *** in vitro and in vivo biological effect has revealed a good biosafety profile and a high tumor suppression *** a novel ND-based system with tumor microenvironment-modulating capability to elevate oxygen content and promote GSH consumption in tumor cells opens new opportunities for enhanced ROS treatment paradigms.

Long‐life lithium batteries enabled by a pseudo‐oversaturated electrolyte

The specific energy of Li metal batteries(LMBs)can be improved by using high‐voltage cathode materials;however,achieving long‐term stable cycling performance in the corresponding system is particularly challenging for the liquid ***,a novel pseudo‐oversaturated electrolyte(POSE)is prepared by introducing 1.1.2,2‐tetrafluoroethyl‐2,2,3,3‐tetrafluoropropyl ether(TTE)to adjust the coordination structure between diglyme(G2)and lithium bis(trifluoromethanesulfonyl)imide(LiTFSI).Surprisingly,although TTE shows little solubility to LiTFSI,the molar ratio between LiTFSI and G2 in the POSE can be increased to 1.1.which is much higher than that of the saturation state,1.*** and experimental results prove that TTE promotes closer contact of the G2 molecular with Li^(+)in the ***,it also participates in the formation of electrolyte/electrode *** electrolyte shows outstanding compatibility with both the Li metal anode and typical high‐voltage ***||Li symmetric cells show a long life of more than 2000 h at 1.mA cm^(−2),1.mAh cm^(−2).In the meantime,Li||LiNi_(0.8)Co_(0.1.Mn_(0.1.O_(2)(NCM81.)cell with the POSE shows a high reversible capacity of 1.4.8 mAh g^(−1.)after 900 cycles at 4.5 V,1.C *** concept of POSE can provide new insight into the Li^(+)solvation structure and in the design of advanced electrolytes for LMBs.

In recent years,multi-modal flexible tactile sensors have become an important direction in the development of electronic skin because of their excellent sensitivity,flexibility and wearable *** this work,a humidity-pressure multi-modal flexible sensor based on polypyrrole(PPy)/Ti_(3)C_(2)T_(x) sensitive film packaged with porous polydimethylsiloxane(PDMS)is investigated by combining the sensitive structure generation mechanism of in situ polymerization to achieve the simultaneous detection of humidity and pressure,which has a sensitivity of 89,1.3.4Ω/%RH in a large humidity range of 0%-97%RH,and response/recovery time of 2.5/1.9 *** tactile pressure sensing has a high sensitivity,a fast response of 67/52 ms,and a wide detection *** device also has excellent performance in terms of stability and repeatability,making it promising for respiratory pattern and motion *** work provides a new solution to address the construction of multi-modal tactile sensors with potential applications in the fields of medical health,epidemic prevention.

Investigation of stress-induced progressive failure of mine pillars using a Voronoi grain-based breakable block model

The Voronoi grain-based breakable block model(VGBBM)based on the combined finite-discrete element method(FDEM)was proposed to explicitly characterize the failure mechanism and predict the deformation behavior of hard-rock mine *** influence of the microscopic parameters on the macroscopic mechanical behavior was investigated using laboratory-scale *** field-scale pillar models(width-to-height,W/H=1.2 and 3)were calibrated based on the empirically predicted stress-strain curves of Creighton mine *** results indicated that as the W/H ratios increased,the VGBBM effectively predicted the transition from strain-softening to pseudo-ductile behavior in pillars,and explicitly captured the separated rock slabs and the V-shaped damage zones on both sides of pillars and conjugate shear bands in core zones of *** volumetric strain field revealed significant compressional deformation in core zones of *** the peak strains of W/H=1.and 2 pillars were relatively consistent,there were significant differences in the strain energy storage and release mechanism.W/H was the primary factor influencing the deformation and strain energy in the pillar *** friction coefficient of the structural plane was also an important factor affecting the pillar strength and the weakest discontinuity *** fracture surface was controlled by the discontinuity angle and the friction *** study demonstrated the capability of the VGBBM in predicting the strengths and deformation behavior of hard-rock pillars in deep mine design.

Tumor progression is closely related to tumor tissue metabolism and reshaping of the microenvironment. Oral squamous cell carcinoma (OSCC), a representative hypoxic tumor, has a heterogeneous internal metabolic environment. To clarify the relationship between different metabolic regions and the tumor immune microenvironment (TME) in OSCC, Single cell (SC) and spatial transcriptomics (ST) sequencing of OSCC tissues were performed. The proportion of TME in the ST data was obtained through SPOTlight deconvolution using SC and GSE1.3322 data. The metabolic activity of each spot was calculated using scMetabolism,and k-means clustering was used to classify all spots into hyper-, normal-, or hypometabolic regions. CD4T cell infiltration and TGF-βexpression is higher in the hypermetabolic regions than in the others. Through CellPhoneDB and NicheNet cell-cell communication analysis, it was found that in the hypermetabolic region, fibroblasts can utilize the lactate produced by glycolysis of epithelial cells to transform into inflammatory cancer-associated fibroblasts (iCAFs), and the increased expression of HIF1. in iCAFs promotes the transcriptional expression of CXCL1.. The secretion of CXCL1. recruits regulatory T cells (Tregs), leading to Treg infiltration and increased TGF-β secretion in the microenvironment and promotes the formation of a tumor immunosuppressive microenvironment. This study delineates the coordinate work axis of epithelial cells-iCAFs-Tregs in OSCC using SC, ST and TCGA bulk data, and highlights potential targets for therapy.

OcBSA:An NGS-based bulk segregant analysis tool for outcross populations

Constructing inbred lines for self-incompatible species and species with long generation times is challenging,making the use of F1.outcross/segregating populations the main strategy for genetic studies of such ***,there is a lack of dedicated algorithms/tools for rapid quantitative trait locus(QTL)mapping using the F1.*** this end,we have designed and developed an algorithm/tool called OcBSA specifically for QTL mapping of F1.*** transforms the four-haplotype inheritance problem from the two heterozygous diploid parents of the F1.population into the two-haplotype inheritance problem common in current genetic studies by removing the two haplotypes from the heterozygous parent that do not contribute to phenotype segregation in the F1.*** of OcBSA on 1.00 simulated F1.populations demonstrated its advantages over other currently available tools in terms of sensitivity and *** addition,the broad applicability of OcBSA was validated by QTL mapping using seven reported F1.populations of apple,pear,peach,citrus,grape,tea,and *** also used OcBSA to map the QTL for flower color in a newly constructed F1.population of potato generated in this *** OcBSA mapping result was verified by the insertion or deletion markers to be consistent with a previously reported locus harboring the ANTHOCYANIN 2 gene,which regulates potato flower *** together,these results highlight the power and broad utility of OcBSA for QTL mapping using F1.populations and thus a great potential for functional gene mining in outcrossing *** ease of use,we have developed both Windows and Linux versions of OcBSA,which are freely available at:https://***/Bioinformaticslab/OcBSA.

Spatial evolution and spatial production of traditional villages from "backward poverty villages" to "ecologically well-off villages": Experiences from the hinterland of national nature reserves in China

With the rapid urbanization process,the space of traditional villages in China is undergoing significant *** the spatial evolution of traditional villages is significant in promoting rural spatial transformation and realizing rural revitalization and sustainable rural *** on the traceability analysis of spatial production theory,this paper constructed an analytical framework for the spatial production evolution of traditional villages,analyzed the spatial evolution process and characteristics of traditional villages by using buffer analysis,spatial syntax,and other research methods,and revealed the characteristics of the spatial production evolution of traditional villages and the driving *** results show that:(1.The village spatial formation and development follow the village life cycle theory and usually develop from embryonic villages to diversified and integrated villages;(2)The evolution of village spatial production is characterized by the diversity of material space,the sublimation of daily life space,and the integration of social system space and generalization of emotional space;(3)The evolution of village spatial production from backward and poor village to ecologically well-off village is influenced by a combination of factors;(4)The village has formed a spatial structure of"people-land-scape-culture-industry",realized comprehensive reconstruction and spatial *** study results reflect the spatial evolution characteristics of traditional villages in mountainous areas in a more comprehensive way,which helps to promote the protection and development of traditional villages in mountainous areas and,to a certain extent,provides a reference for the development of rural revitalization.

MODIFIED STOCHASTIC EXTRAGRADIENT METHODS FOR STOCHASTIC VARIATIONAL INEQUALITY

In this paper,we consider two kinds of extragradient methods to solve the pseudo-monotone stochastic variational inequality ***,we present the modified stochastic extragradient method with constant step-size(MSEGMC)and prove the convergence of *** the strong pseudo-monotone operator and the exponentially growing sample sequences,we establish the R-linear convergence rate in terms of the mean natural residual and the oracle complexity O(1.ǫ).Second,we propose a modified stochastic extragradient method with adaptive step-size(MSEGMA).In addition,the step-size of MSEGMA does not depend on the Lipschitz constant and without any line-search ***,we use some numerical experiments to verify the effectiveness of the two algorithms.
