Cand2 inhibits CRL-mediated ubiquitination and suppresses autophagy to facilitate pathogenicity of phytopathogenic fungi

The ubiquitin–proteasome system and the autophagy system are the two primary mechanisms used by eukaryotes to maintain protein homeostasis,and both are closely related to the pathogenicity of the rice blast *** this research,we identified MoCand2 as an inhibitor of ubiquitination in Magnaporthe *** this role,MoCand2 participates in the regulation of autophagy and ***-cifically,we found that deletion of MoCand2 increased the ubiquitination level in ***,whereas overexpression of MoCand2 inhibited the accumulation of ubiquitinated *** analyses showed that MoCand2 is a subunit of Cullin-RING ligases(CRLs).It suppresses ubiquitination by blocking the assembly of CRLs and downregulating the expression of key CRL *** research indi-cated that MoCand2 regulates autophagy through ***2 knockout led to over-ubiquitination and over-degradation of MoTor,and we confirmed that MoTor content was negatively correlated with autophagy *** addition,MoCand2 knockout accelerated the K63 ubiquitination of MoAtg6 and strengthened the assembly and activity of the phosphatidylinositol-3-kinase class 3 complex,thus enhancing *** ubiquitination and autophagy in DMocand2 resulted in defects in growth,conidiation,stress resistance,and ***,sequence alignment and functional an-alyses in other phytopathogenic fungi confirmed the high conservation of fungal *** research thus reveals a novel mechanism by which ubiquitination regulates autophagy and pathogenicity in phyto-pathogenic fungi.

Curcumin Interferes with TGF-β1-Induced Fibrosis in NRK-49F Cells by Reversing ADAMTS18 Gene Methylation

Objective To explore the molecular mechanism by which curcumin affects renal interstitial fibrosis(RIF)progression by regulating ADAM metallopeptidase with thrombospondin type 1 motif 18(ADAMTS18)*** NRK-49F cells RIF model were induced with transforming growth factorβ1(TGF-β1).Effects of different concentrations of curcumin(0,10,20,and 30μmol/L)on cell proliferation,cell cycle,cell apoptosis as well as cyclin D1 expression were analyzed by cell counting kit-8,flow cytometry and Western blot,***18 methylation levels were determined by methylation-specific polymerase chain ***18,fibronectin(FN),type I collagen(Col-I)and alpha-smooth muscle actin(α-SMA)mRNA and protein expressions were analyzed by real-time PCR(RT-PCR)and Western blot,***,cells were treated with 50 mmol/L 5-aza-2′-deoxycytidine(5-aza-dC,demethylation agent)for 72 *** of curcumin on extracellular matrix(ECM)deposition was evaluated by immunochemical staining and Western ***-49F cells were transfected with ADAMTS18 small interfering RNA and grouped into a normal control,ADAMTS18-knock-out(KO),and ADAMTS18-KO+30μmol/L curcumin groups,and whether curcumin can reverse the effect of ADAMTS18 knockdown on RIF was *** Compared with the control group,TGF-β1 significantly inhibited the proliferation of NRK-49F cells,blocked the G1/G0 phase,promoted cell apoptosis and inhibited cyclin D1 expression(P<0.01).Among the different concentrations of curcumin,30μmol/L curcumin significantly reversed these processes(P<0.01).Immunochemical staining and Western blot results showed that curcumin significantly inhibited the deposition of FN,Col-I andα-SMA(P<0.01).Curcumin and 5-zaz-dC had synergistic effects,promoting ADAMTS18 expression,removing ADAMTS18 methylation,and reducing ECM ***18 knockdown promoted ECM accumulation,and curcumin reversed this process(P<0.01).Conclusion TGF-β1-induced fibrosis in NRK-49F ***

Color-polarization synergistic target detection method considering shadow interference

Aiming at the problem of shadow interference in UAV's ground reconnaissance,a color and polarization synergistic target detection method is proposed for anti-shadow interference,based on the influence of two physical characteristics(wavelength and polarization)under different illuminations.A valid fusion strategy is proposed via integrating two separate detection results on color and polarization ***,a local enhancement and recognition module(LER)is introduced to boost the *** on our built dataset,experimental results show that our method achieves mAPof 87.76%,and12.37%higher than that of color image and 14.68%higher than that of polarization image.

With the deepening of neural network research,object detection has been developed rapidly in recent years,and video object detection methods have gradually attracted the attention of scholars,especially frameworks including multiple object tracking and *** current works prefer to build the paradigm for multiple object tracking and detection by multi-task *** with others,a multi-level temporal feature fusion structure is proposed in this paper to improve the performance of framework by utilizing the constraint of video temporal *** training the temporal network end-to-end,a feature exchange training strategy is put forward for training the temporal feature fusion structure *** proposed method is tested on several acknowledged benchmarks,and encouraging results are obtained compared with the famous joint detection and tracking *** ablation experiment answers the problem of a good position for temporal feature fusion.

The long chain branching(LCB)polyglycolide(PGA)was successfully prepared by the successive reactions of the terminal hydroxyl groups of PGA with triglycidyl isocyanurate(TGIC)and pyromellitic dianhydride(PMDA).The influence of LCB produced by functional group reaction on rheological and crystallization behavior was studied and discussed through linear rheology,uniaxial elongation and DSC(differential scanning calorimetry).The much higher viscosity and the more notable strain hardening behavior of modified PGA indicates the LCB with high degree of entanglements are *** melt strength of PGA is finally improved greatly and can make su re that the supercritical CO_(2)foaming can be carried out successfully.

Neuroinflammation exacerbates secondary damage after spinal cord injury,while microglia/macrophage pyroptosis is important to *** RNAs(circRNAs)play a role in the central nervous ***,the functional role and mechanism of circRNAs in regulating microglia/macrophage pyroptosis after spinal cord injury are still poorly *** the present study,we detected microglia/macrophage pyroptosis in a female rat model of spinal cord injury,along with upregulated levels of circ0000381 in the spinal *** further experimental results suggest that circ0000381 may function as a sponge to sequester endogenous microRNA423-3p(miR-423-3p),which can increase the expression of NOD-like receptor 3(NLRP3),a pyroptosis ***,upregulation of circ0000381 may be a compensatory change after spinal cord injury to attenuate microglia/macrophage ***,knockdown of circ0000381 expression exacerbated microglia/macrophage ***,our findings provide novel evidence for the upregulation of circ0000381,which may serve as a neuroprotective mechanism to attenuate microglia/macrophage pyroptosis after spinal cord ***,circ0000381 may be a novel therapeutic target for the treatment of spinal cord injury.

Nitrosamines are a class of carcinogens which have been detected widely in food,water,some pharmaceuticals as well as *** objectives of this paper include reviewing the basic information on tobacco consumption and nitrosamine contents,and assessing the health risks of tobacco nitrosamines exposure to Chinese *** searched the publications in English from“Web of Science”and those in Chinese from the“China National Knowledge Infrastructure”in 2022 and collected 151 literatures with valid *** content of main nitrosamines in tobacco,including 4-(methylnitrosoamino)-1-(3-pyridyl)-1-butanone(NNK),N-nitrosonornicotine(NNN),N-nitrosoanatabine(NAT),N-nitrosoanabasine(NAB),total tobacco-specific nitrosamines(TSNA),and N-nitrosodimethylamine(NDMA)were *** information of daily tobacco consumption of smokers in 30 provinces of China was also ***,the intakes of NNN,NNK,NAT,NAB,TSNAs,and NDMA via tobacco smoke were estimated as 1534ng/day,591 ng/day,685 ng/day,81 ng/day,2543 ng/day,and 484 ng/day by adult smokers in30 provinces,*** cancer risk(CR)values for NNN and NNK inhalation intake were further calculated as 1.44×10^(-5)and 1.95×10^(-4).The CR value for NDMA intake via tobacco smoke(inhalation:1.66×10^(-4))indicates that NDMA is similarly dangerous in tobacco smoke when compared with the *** China,the CR values caused by average nitrosamines intake via various exposures and their order can be estimated as the following:smoke(3.75×10^(-4))>food(1.74×10^(-4))>drinking water(1.38×10^(-5)).Smokers in China averagely suffer 200%of extra cancer risk caused by nitrosamines in tobacco when compared with non-smokers.

The circular electron-positron collider(CEPC)is designed to precisely measure the properties of the Higgs boson,study electroweak interactions at the Z-boson peak,and search for new physics beyond the Standard *** a component of the 4th conceptual CEPC detector,the drift chamber facilitates the measurement of charged *** study implemented a Geant4-based simulation and track reconstruction for the drift *** the simulation,detector construction and response were implemented and added to the CEPC simulation *** development of track reconstruction involves track finding using the combinatorial Kalman filter method and track fitting using the tool of *** the simulated data,the tracking performance was *** results showed that both the reconstruction resolution and tracking efficiency satisfied the requirements of the CEPC experiment.

Edible prodelphinidins that improve postprandial hyperglycemia were first discovered in Chinese bayberry(Myrica rubra Sieb. et Zucc.) fruits

Plant polyphenols are potential substitutes for clinical α-glucosidase inhibitors. Our previous studies indicated that prodelphinidins from Chinese bayberry leaves (BLPs) have stronger α-glucosidase inhibitory activity than other common polyphenols， but they have no safe history of consumption. There is a reasonable prospect that prodelphinidins from Chinese bayberry fruits (BFPs) can improve postprandial hyperglycemia， though known active components are only myricetin and cyanidin-3-O-glucoside. Hence， the aim of this study was to analyze structure and hypoglycemic effect of BFPs of “Biqi” (BQPs) and “Dongkui” (DKPs) cultivars， and compare their difference with BLPs. The contents of BQPs and DKPs were (221.73±0.60) and (136.82±4.33) mg EC equivalents/g dry weight， respectively. The most subunits were EGCG along with a small amount of EGC， ECG and EC connected by B-type linkages and a small portion of A-type linkages with mean polymerization degree of 4.25 and 4.08， respectively. Importantly， BQPs and DKPs were α-glucosidase inhibitors with IC50of 11.91 and 9.47 μg/mL respectively， which were significantly stronger than BLPs. DKPs could also improve postprandial hyperglycemia of normal mice and high fat diet-induced type Ⅱ diabetic mice. Therefore， edible prodelphinidins， which have stronger hypoglycemic effect than BLPs， were first found in Chinese bayberry fruits.
