Differential expression patterns and functions of IFNAR1 and IFNAR2 in co-culture model of bovine blastocysts and endometrial cells

摘 要：Objective:Interferon-tau(IFNτ),a type I interferon,is an antiluteolytic factor secreted by trophoderm during the pregnancy of ***-τ transmits signals or stimulates the expressions of some factors to build maternal recognition and keep pregnancy by binding its receptors,IFNτ receptor 1(IFNAR1) and IFNτ receptor 2(IFNAR2).Up to now,the IFNτ-IFNAR cascade regulation process between the embryo and uterine epithelial cells has not been clearly *** study aimed to explore the functions of IFNAR in regulating the molecular mechanism of precise embryo-uterine ***:the technologies of in vitro fertilization,parthenogenetic activation and somatic cell nuclear transfer were used in this study to obtain these in vitro produced blastocysts and in vivo ones to compare the differences of their own IFNτ secretion and their in vitro co-culture effect with endometrial epithelial ***:In this study,we found that the expression and location of IFNτ in blastocysts from different *** addition,IFNτ,IFNAR1 and IFNAR2 are all located in the trophoblast cells of the ***,the fluorescence intensity of IFNAR1 is consistent with IFNτ.The co-culture of bovine blastocysts and their endometrial epithelial cells from different sources stimulated the expressions of IFNAR1,IFNAR2 and downstream Integrin αvβ3,WNT7 A,*** the expression of IFNAR1 and IFNAR2 in embryos and co-culture with EEC also reduced the expressions of Integrin αvβ3,WNT7 A,ISG15 in *** out IFNAR1 and IFNAR2 with CRISPER-cas9,co-cultivation with EEC found that although both can reduce the expression of Integrin αvβ3 and WNT7 A in EEC,the deletion of IFNAR2 gene has a greater impact than the deletion of IFNAR1 *** IFNAR1-/IFNAR2-co-cultured with IVF and PA embryos,the expression of Integrin αvβ3 was inhibited,while the expression of WNT7 A was not inhibited in IVF ***1-/IFNAR2+and IFNAR1/IFNAR2were co-cultured with IVF,the expression of
