摘      要:Objective:To investigate the role of H19 on pituitary adenomas and its underlying mechanism in vitro and in vivo..Methods:Pituitary tumor tissues were obtained from pituitary tumor patients who underwent surgery between 2015 and 2020 at the Department of Neurosurgery,Ruijin Hospital of Shanghai Jiaotong University and Department of Neurosurgery,First Affiliated Hospital of Wenzhou Medical University;The Cell Counting Kit-8(CCK-8) assay was used to test cell viability;Real-time quantitative PCR was used to test the level of H19 and miR-93a;Western blot experiments were conducted to test the ATG7 expression level;Luciferase reporter assay and RNA immunoprecipitation were used to ensure the combination of H19 and miR-93a;Xenograft experiments were used to test the effect of H19 and miR-93a in ***:H19 inhibited tumor growth and had a synergistic effect with DA treatment on prolactinomas in vitro and in ***,H19 promoted ATG7 expression in pituitary tumor cells by inhibiting miR-93a *** addition,a potential binding site between miR-93 and H19 was confirmed,and low expression of miR-93 was previously found in DA-resistant ***,we showed that miR-93a regulates ATG7 expression by targeting ATG7 ***:Our study has identified the role of the H19-miR-93-ATG7 axis in tumor growth and DA treatment of pituitary tumors,which may be a potential therapeutic target for human pituitary tumors..
