CD4+T细胞来源的白介素-21加重小鼠脊髓损伤(英文)

摘 要:Immune responses and neuroinflammation are critically involved in spinal cord injury(SCI).In this work,we examined the CSF of SCI patients and found a significantly increase in interleukin-21(IL-21).Similaily,the levels of IL-21 in SCI mice increased obviously.Depletion of IL-21 receptor or IL-21 resulted in improved recovery after SCI.On the other hand,after SCI,CD4+T cells expressing IL-21 were infiltrated into the lesion sites within 24 hrs post-injury.Depleting CD4+T cells similarly contributed to functional recovery.Furthermore,M1 macrophages promote inflammatory responses in SCI.The results showed that macrophages expressed IL-21 r,and the percentage of M1 macrophages was notably decreased in IL-21 r and IL-21 mice after SCI.In conclusion,CD4T cell-derived IL-21 aggravates lesions in SCI,by promoting the differentiation of M1 macrophages.Thus,regulating the levels of IL-21 may be a potential approach of biointervention after SCI.
