The effect of bufalin in human esophageal squamous cell carcinoma cells and xenograft model of nude mice

摘 要:Background: Bufalin exhibits significant antineoplastic activities in many neoplasms. The aim of this study was to evaluate the effects of Bufalin on ERK/RSK2 pathway and EMT and their mechanisms of action in human esophageal squamous cell carcinoma cells(ECA109 cells). Methods: ECA109 cells were incubated with bufalin, TUNEL, Transwell migration/invasion assay, PT-PCR and Western blotting were performed in ECA109 cell lines. Established the ESCC transplantation neoplasm in nude mice and treated with Bufalin. The mRNA levels(ERK, RSK2, E-Cadherin and Vimentin) and the protein levels(ERK1/2, pERK1/2, RSK2, pRSK2, BAD, pBAD and Fascin-1) were detected by Real-time quantitative PCR and Western blot analysis in nude mice transplanted neoplasm. Results: Bufalin inhibited ECA109 cell migration, invasion and promote apoptosis. Treatment with Bufalin significantly decreased relative expression of ERK, RSK2 and FAK m RNA in nude mice *** protein levels of pERK, pRSK2, pBAD and Fascin-1 was ***: Bufalin can exert an inhibitory effect on esophageal cancer ECA109 cells and xenografts in nude mice through the ERK/RSK2 pathway and also significantly inhibit FAK and the process of EMT in esophageal cancer.
