Expression profiles of circRNAs and mRNAs in early brain injury following experimental subarachnoid hemorrhage

摘      要:Circular RNAs(circRNAs) are a special novel type of a stable, diverse and conserved noncoding RNA in the mammalian brain, well conserved in sequence and involved in many nervous system diseases. This study aims to reveal the expression profiles of circRNAs and mRNAs in subarachnoid hemorrhage(SAH) brain tissue of mice by high-throughput sequencing. Male C57BL/6J mice were used to construct the SAH model. At 24 h post-SAH, the cortex around perforation site was collected. The total RNA was extracted to construct the cDNA library. High-throughput sequencing and bioinformatics analysis were carried out. The results showed that a total of 26 circRNA transcripts and 804 mRNA transcripts were aberrantly expressed after SAH. RT-qPCR further validated the gene expression. Bioinformatics predicted that these 26 circRNAs contain binding sites for many microRNA. Gene ontology(GO) enrichment and Kyoto Encyclopedia of Genes and genomes(KEGG) analysis showed that most mRNAs originated from the parent genes of circRNAs are involved in axon guidance, inflammatory and immune response, protein binding and other pathophysiological process. In conclusion, in mice with SAH, the circRNA profile in brain tissue differs markedly from that in normal brains. These changes could be associated to the possible functional implication to post-SAH pathophysiology.
