Whole genome re-sequencing of Eimeria tenella reveals positive selection in the precocious lines

摘      要:Chicken coccidiosis,caused by Eimeria species,leads to huge economic losses in poultry *** immunization with attenuated live vaccine seems to be the most cost-effective way for controlling this *** produce such attenuated live vaccine,different Eimeria species/strains were selected for precocious lines,which characterized by greatly shorted prepatent time and faster endogenous *** these precocious line-based vaccines are widely used,the genetic mechanisms of the precocity phenomenon remain *** this study,we isolated 19 *** strains from different geographical samples by multiple-oocyst *** the determination of the prepatent time,oocyst output curve,total oocyst output and damage to the chicken intestine(lesion scores) for each strain,5 low-virulent strains(prepatent time 120 h,mean lesion score(1× 104 inoculation) 2) and 14 virulent strains were *** DNA extracted from each strain was used for Illumina sequencing.A total of 112 Gb clean data were produced and the mean coverage was 106×,and 679318 SNPs were detected in *** linkage disequilibrium decay distance of the virulent group was much shorter than the lowvirulent group,which means the low-virulent precocious lines undergo strong positive ***,we found a large select sweep region(260 Kb) in Lachesisroup 9(chromosome in new reference genome) by fixation index(FST) in 10 kb sliding window with a step size of 1 *** protein coding genes under positive selection is under *** results open a window for deciphering the mechanisms of the precocity in Eimeria.
