摘 要:Our laboratory has developed a continuous manufacturing platform for liposomes using a co-axial jet in co-flow and incorporating online PAT(process analytical technology). DOE(design-of-experiments) approaches have been applied in the system design. The platform development will be presented along with application to liposomal drug delivery systems. Our continuous manufacturing platform is capable of tuning liposome particle size with precise control of the polydispersity index(PDI) to achieve monodisperse product. Identifying key process parameters and material attributes of liposomal formulations, such as the z-average particle size, polydispersity index and zeta-potential, are extremely important since they may impact the formulation quality, drug loading and in vivo performance of the final liposomal drug product.
