F-box like motif of CCYV P22 is crucial for silencing suppressor activity by interacting with CsSkp1LB1

摘 要:Plants employ RNA silencing as a virus defense *** response,viruses encode various RNA silencing suppressors to counteract the antiviral ***,we identify P22 as a silencing suppressor of CCYV and show that P22 interacts with Cucurmis sativus orthologs of S-phase kinase related protein 1(SKP1) via its F-box like *** F-box like motif deletion mutant of P22 abolished P22 pathogenicity and silencing suppressor *** analysis of Nicotiana benthamiana leaves expressing P22 and its F-box like motif deletion mutant further confirmed indispensibilty of F-box like motif for the ubiquitin mediated proteolysis and methionine metabolism.S-adenosyl--methionine synthase(SAMS),a key enzyme for HEN1 activity,was undetectable in P22 expressing leaves while expressed in its deletion *** together,our results suggest that P22 acts as silencing suppressor possibly via depriving HEN1 of its substrate S-adenosyl--methionine(SAM) through the ubiquitin mediated proteolysis.
