摘      要:Embryo dormancy(embryo diapause) is a reproductive strategy used by approximately 100 different mammals to avoid the risk of exposing their offsprings to unfavourable environmental conditions by delaying embryo *** molecular mechanism of embryonic diapause is not yet *** hypothesize that endometrial exosome/microvesicles(EV) regulates embryo *** detected miRNA profiles in EV between dormant and activated uterine fluid and found that the members of let-7 family were highly expressed in dormant uterine fluid ***,embryos have ability to take up endometrial exosomes/*** examine the roles of let-7 in embryo dormancy,we eletroporated let-7 a into embryos and found that embryos electroporated with let-7 can be alive even after the culture for up to Day 13.18.8%of embryos from let-7 g transgenic mice developed to term after culture for 6 days(ie Day 10).Microarray analysis showed that 47%gene expression were similar between let-7-treated and P4-induced dormant embryos compared to D4 or E2-activated embryos and most of genes were related to cell cycle and *** results showed that let-7 has an important role in the process of embryo dormancy in *** level of let-7 was elevated in Iin28 transgenic embryos which were transferred into wild-type delayed implantation model for 3 *** result suggests that the increased levels of let-7 in dormant embryo were mainly derived from outside *** exosomes/microvesicle of highly expressed let-7 prolonged the survival time of embryos in *** conclusion,our results demonstrated that let-7 of endometrial exosomes/microvesicles regulated embryo dormancy in mice.
