Identification of γδ T cells from zebrafish provides insights into their important role in adaptive humoral immunity

摘 要:The γδ T subset is an evolutionarily primitive T cell population characterized by distinct TCRs and functional roles in the innate and adaptive *** has been well documented in humans and mouse ***,little is known about the occurrence and existence of this T cell subset in ancient *** the present study,we report the molecular and cellular characterizations of γδ T cells from a zebrafish(Danio rerio) *** constant regions of zebrafish TCR-γ(DrTRGC) and TCR-8(DrTRDC) were initially *** γδ T cells accounted for 7.7% to 20.5%of the total lymphocytes in spleen,kidney,peripheral blood,skin,gill and intestine *** possess typical morphological features of lymphocytes with a surface phenotype of γδ*** γδ T cells functionally showed a potent phagocytic ability to both soluble and particulate *** can act as an antigen presenting cell(APC)to initiate the antigen(KLH)-specific CD4 T cell activation and to induce the subsequent B cell proliferation and antibody(IgM) ***,zebrafish γδ T cells also play a critical role in fish specific IgZ production in intestinal *** findings indicated that γδ T cells had been originated as early as teleost fish,which providing valuable insights into the evolutionary history of T cell *** hope that this study would be used as a guide to develop a zebrafish model for the cross-species investigation of γδ T cell biology.
