摘      要:We have previously demonstrated the improving role of hydrogen sulfide(HS),the third gasotransmitter,in the cognitive function of streptozotocin(STZ)-induced diabetic *** has been confirmed that impaired hippocampal autophagic flux has a key role in the pathogenesis of cognitive impairment and that ornithine decarboxylase(ODC)/spermidine(Spd) pathway plays an important role in the formation of memory by promoting autophagic ***,we investigated the roles of hippocampal ODC/Spd pathway and autophagic flux in HS-attenuated cognitive impairment in streptozotocin(STZ)-induced diabetic *** this study,we found that sodium hydrosulfide(NaHS,a donor of HS) up-regulated the expression of ODC and the level of Spd in the hippocampus of STZ-induced diabetic *** also found that NaHS markedly improved the autophagic flux in the hippocampus of STZ-exposed rats,as evidenced by the increases in the numbers of autophagosomes as wells as the expressions of LC3-Ⅱ,Beclin-1,and P62 in the hippocampus of cotreatment with NaHS and STZ ***,difluoromethylomithine(DFMO,an inhibitor of ODC) markedly prevented the up-regulated role of NaHS in the level of hippocampal Spd and reversed the protection of NaHS against the impairments in hippocampal autophagic flux as well as cognitive function in STZ-exposed *** findings indicated that improving hippocampal autophagic flux plays a key role in HS-attenuated cognitive impairment in STZ-induced diabetic rats,which is mediated by up-regulation of hippocampal ODC/Spd pathway.
