摘      要:Objective X/A like cells which secret ghrelin/nesfatin-1 arc endocrine cells mainly located in the gastric.We previously found that ghrelin/nesfatin-1 regulated glucose metabolism and Mechanistic target of rapamycin(mTOR)regulated the secretion of ghrelin/nesfatin-1.However whether mTOR signals in X/A like cells regulate glucose metabolism remains unclear.Methods:Ghrl-TSCl-/-mice and Ghrl-mTOR-/-mice were used to activate or knock out mTOR in X/A like cells.The expression of mTOR signals was investigated by western blotting.OGTT/ITT were conducted when mice were 8/12/16 weeks old.Insulin expression was delected by Real-Time PCR and western blotting.Plasma insulin was detected by enzyme immunoassay.Results:(1)mTOR signals were specifically intervened in X/A like cells.(2)The glucose tolerance was exacerbated in Ghrl-TSC1-/-mice.(3)The expression of insulin and plasma insulin were both reduced in Ghrl-TSC1-/-mice.Conclusion:mTOR signals in X/A like cells regulate glucose metabolism.
