To expand allo-specific Tscm by suppression of T-cell differentiation in a co-culture in vitro

摘 要：Background/Aims:Immunotherapy based on adoptive transfer of T cells is hampered by the lack of persistent and antigen-specific human T *** stem cells which possess capacity of self-renewal and multipotency could provide an good sources of T *** we explored a method to expand allo-specific Tscm in ***:To prepare allo-restricted Tscm,an in vitro co-culture of LCL cell line with allogeneic peripheral blood mononuclear cells(PBMC) was established in the presence of *** phenotypes of the prepared Tscm were detected by FCM with corresponding *** proliferating Tscm were sorted by multicolor FACS on the seventh day after the *** Tscm were restimulated with the LCL for 24 h to analyze the differentiation and ***,the allo-specificity of the Tscm was checked by an HLA-mismatched LCL during the ***:After the coculturing,the sorted cells displayed a Tscm phenotype CD3CD45 RACD62 LCD95CCR7CD122.65%Tscm restimulated for 24 h by the cognate LCL were able to differentiate into Tem and Tef,and 15%Tscm maintained the phenotype mentioned ***,there was no differentiation when restimulated by an HLAmismatched LCL,indicating the alloantigen-specificity of the prepared *** the differentiation of the Tscm,the incubated cell bulks showed alloantigen-specific cytotoxicity and cytokine ***:This study provided a practical method for allo-Tscm preparation,the prepared Tscm showed the stem cell properties,differentiation and *** importantly,the generated Tscm shows alloantigen-specificity.
