Effects of Kuntai Capsule on follicular development and oocyte cohesin levels in aged mice

摘 要:Women with advanced maternal age experience more incidences of sterility, miscarriage and embryo aneuploidy. Recent studies have revealed that age-related aneuploidy is mainly caused by the reduction of cohesin complexesin oocyte. Kuntai Capsule is a kind of Traditional Chinese medicine that was firstly used to retrieve the menopausal syndrome. Several studies have indicated its effect to improve the ovarian reserve and oocyte quality in patients undergoing in vitro fertilization-embryo transfer. To evaluate whether Kuntai Capsule could affect cohesion levels and improve the ovarian function of aged mice, 9 month-old C57 BL/6 J mice were divided into different groups and treated with saline, 17β-estradiol, and different concentrations of Kuntai Capsule for 4 weeks. HE staining and anti-VASA staining were used to detect the amounts of follicles. The apoptosis of follicles was measured by anti-γH2 AX staining and TUNEL assay. The density of cohesin subunits-REC8, SMC1β and SMC3 in oocytes were evaluated by immunofluorescent staining. Our findings revealed that administration of Kuntai Capsule increases the total number as well as the number of primordial and primary follicles in a dose-dependent manner. Outcome fromanti-γH2 AX staining and TUNEL assay demonstrated that Kuntai Capsule and E2 partly suppress the apoptosis of follicles. Furthermore, it showed an increased trend of the meiotic-specific cohesin subunits REC8 and SMC1βafter administration with E2 and Kuntai Capsule, and no obvious change in the level of cohesin subunit common to mitotic *** present results revealed that supplement of Kuntai Capsule in aged mice could enhance the number of follicles, suppress apoptosis of oocytes and have a trend to elevate the meiotic-specific cohesin subunits REC8 and SMC1βin oocytes, which implies a beneficial effect on the ovarian function in terms of the quantity and quality of follicles.
