A Pathogenic Fungus Phoma *** Identified from Tea Leaf Spot Disease in Shiqian County,Guizhou Province,China

摘 要:The tea leaf spot disease is always taken place in the region with the characteristics of the low-temperature and high-humidity containing with Shiqian County,Guizhou *** disease always infects the tender leaves and shoots,which lead to the lowest tea quality and the significant production *** identify the pathogen of the tea spot disease,this work isolated and identified the pathogen of this disease from the tea leaves,and determined the pathogenicity of the pathogen according to Koch’s *** study indicated that the isolated strain of GZSQ-4 represent the features as following;on PDA medium,margin regular of the colonies,aerial mycelium closely growing,white mycelium in the early stage;then gradually change to grey brown,black brown in the center,with white margin in the latter stage;reverse brown to black brown near the center,with the white *** OA medium,margin regular of the colonies,aerial mycelium woolly,pale brown to brown near the center,and produce a lot of black *** the optical microscope and SEM indicated that the septate mycelium contains the inclusion granule;Pycnidia represent subglobose,glabrous,thin-walled,solitary or confluent,50 190μm,1 or 2 ostioes with a neck;Conidia represent ellipsoidal or ovoid,thin-walled,smooth,hyaline,aseptate,(2) μm×(1 .5) μm,with 0 guttules in two polar *** genes of ITS,β-tubulin and LSU were further amplified,purified and sequenced,then the phylogenetic trees were constructed using MEGA6.0 software and Neighbor-joining *** results indicated that the strainof GZSQ-4 was closely related to the strains of Phoma ***-2014 b(LC 1633) and Didymel-1 a bellidis(CBS 714.85).Integrated with the above results,the pathogen isolated by our research group are preliminarily identified to Phoma sp..
