WW domain-containing protein genes identification in the silkworm and expansion in eukaryotes

摘      要:WW domain-containing proteins(WWCPs) have been implicated in a variety of cellular processes, such as development,differentiation and cellular homeostasis, take critical roles in major human diseases including cancer as well as in major signaling cascades, making them targets for new diagnostics and therapeutics. Although there are a few studies on identification and function of WWCPs, it’s never having been systematically investigated in *** this study, by searching the silkworm genome assembly with the HMM model of WW domain(PF00397), 24 proteins in the silkworm with 39 WW domains have been identified totally. Fifteen silkworm WW domain could be classified into Group I, another fifteen WW domains constituted the Group Ⅱ/Ⅲ(figure1b), two WW Domains belong to the Group Ⅳ, and nine WW domain derived from eight silkworm WWCPs could not been classified into any group. The WWCPs of silkworm should take variety roles as RNA transcription and processing, cell apoptosis and differentiation, ubiquitination, control of the cytoskeleton and cell signal transduction based on the gene *** further analyzed the potential WWCP binding proteins in silkworm with regular expression of PPx Y motif(PY motif) and poly-proline motif. We found that nearly eighteen percent of silkworm proteins contain PPx Y motif and thirty percent proteins totally in 14,623 predicted gene models of silkworm contain variety motifs that could be recognized/bond by WW domain potentially. GO analysis revealed that dozens of WW domain binding proteins we identified take part in Wnt, Hedgehog, Notch, m TOR, EGF and Jak-STAT signaling pathway besides the other various biology process include nutrient metabolism. The tissue expression patterns of WWCP genes and WWCP potential-binding protein genes on day 3 of the fifth instar were also examined by gene microarray analysis, and some of these genes shown tissue or sex-dependent expression pattern in ***, we identified and analyzed
