An in vitro model of oligodendrocyte precursor cells differentiation and development

摘 要:Objective It is now widely accepted that polydendrocytes are oligodendrocyte progenitor cells(OPCs) that generate oligodendrocytes in the developing and mature CNS and serve as the primary source of remyelinating cells in demyelinated ***,OPC has been identified that it can fire action potentials,and their excitation is driven by synapses from *** study aims to provide an in vitro model of OPCs differentiation,by which we could further demonstrate the mechanisms of factors effect the survival and differentiation of oligodendrocytes at the level of morphological, myelin protein as well as ion current *** OPCs were obtained from E15 rat spinal cords and were stimulated to differentiate into more mature oligodendrocytes in the presence of 3,3’,5-triiodo-L-thyronin sodium salt(T3). After T3 introduction,OPCs were induced to differentiate at various time points(2d,4d,6d and 8d) in *** staining with differentiating oligodendrocyte markers,A2B5,O4,O1,receptor-interacting protein(RIP) and myelin basic protein(MBP) was used to determine the developmental stage of ***(1) Proliferating OPCs were expanded through multiple passages in *** than 94%of OPCs expressed A2B5,an oligodendrocyte progenitor marker;about 10%OPCs expressed 04;and few OPCs expressed O1,RIP or MBP.(2) After T3 induction at various time points in vitro,A2B5-expressing OPCs gradually decreased to 27.0%at 8 d,and differentiated into preoligodendrocytes (O4,70.1%at 6d),immature oligodendrocytes(O1:46.9%,RIP:30.1%at day 6) and mature oligodendrocytes (MBP,21.4%at 6d).(3) The percentage of GFAP-immunoreactive cells increased from 3.3%at Od to 49.9%at 8d. Conclusion In vitro environment that mimics oligodendrocyte differentiation can be successfully achieved.
