The Effects of Self-assembled IKVAV peptide nanofibers on Dorsal Root Ganglion Neurons

摘 要:Objective To observe the effects of self-assembled IKVAV peptide nanofibers on dorsal root ganglia neurons(DRGn)through co-culture of DRGn and *** The peptide was dissolved in NaOH solution where PH was equal to 8.5 and the content of peptide was 1wt%.1wt% peptide covered with coverslip was triggered to be self-organized into gel by the addition of DMEM/*** was detected by *** root ganglia were collected from newborn rats,and then the tissue were dispersed into individual cell suspension by primary isolated culture *** the cell suspension was divided into experimental group and control group. In experimental groups(EG):DRGn were transplanted on the surface of 2-D *** control groups(CG):DRGn were transplanted on the polylysine. Growth of DRGn was observed by ***’s live time and the length of neurite in EG and CG were observed with cell counting and immunohistochemistry methods. Results TEM showed that the self-assembly gel was composed of network *** length of neurite of DRGn in EG and CG is(43.8 ±10.4)μm、(33.4±5.75)μ*** neuron percentage after 14 days in EG and CG is(43.60±4.83)%、 (39.04±3.87)%.Conclusion Self-assembled IKVAV peptide nanofibers,which can prolong the live time of neurons and promote neurite sprouting and direct neurite growth,is a bioactivity scaffold,can be acted as a superordinary nerve tissue engineering scaffold.
