I3C and DIM induce apoptosis of SMMC 7721 and HepG2 hepatic tumor cells by down-regulating survivin expression

摘      要:Background The indole derivatives indole-3-carbinol(I3C) and 3,3 -diindolylmethane(DIM) have been purported to be chemopreventive for certain ***,there are limited data regarding the effects of I3C and DIM on hepatic tumor cells and their mechanism of action remains to be ***,in this study,using two distinct hepatic tumor cell lines,SMMC 7721 and HepG2,we determined the in vitro effect of I3C and DIM on cell proliferation,apoptosis,mitochondrial function and the expression of proteins involved in these processes. Results I3C and DIM produced a significant decrease in the proliferation of SMMC 7721 and HepG2 cells and a significant increase in their rate of apoptosis in a time-and concentration-dependent *** inhibition of survivin expression by I3C and DIM produced an arrest in the cell cycle at the G0/G1 ***, I3C and DIM produced an activation of mitochondrial signaling,accompanied by a decrease in the mitochondrial membrane potential,cytochrome c release and the activation of caspase-9 and caspase-3. Conclusions Overall,these findings indicate that I3C and DIM significantly inhibited the proliferation of SMMC 7721 and *** could occur via a number of mechanisms,including the inhibition of survivin expression,as well as the activation of apoptosis.
