Cloning and comparative study of trehalose-6-phosphate synthase genes in seaweed

摘 要:Trchalose is a non-reducing disaccharide of two glucose units that functions as a stress protection metabolite in the stabilization of biological structures under stress tolerance in various *** biosynthesis of trehalosc is catalyzed by a two-step process that involves trehalosc-6-phosphate synthasc(TPS)and trchalosc-6-phosphate phosphatase(TPP).In this study,the full-length cDNA sequence(3219 bp)of the trehalose-6-phosphate synthase gene of Porphyra yezoensis(PyTPS)was isolated by RACE(rapid amplification of eDNA ends)-PCR. PyTPS encodes the TPS protein of 908 amino acids with a calculated molecular mass of 101786 Daltons,consisting of a TPS domain at the N-terminus and a putative TPP domain at the *** putative TPS sequence was deposited in GenBank(NCBI)with the accession numbers AY729671(mRNA)and AAW27916(protein).The homology alignment for the TPS proteins from five different organisms indicates that the most closely related sequences to PyTPS were those from OsTPS and AtTPS,whereas the most distant sequence to PyTPS was the one from EcOtsAB.
