摘      要:正At present our understanding of the molecular mechanisms underlying the bone marrow stromal cells(BMSCs) differentiating into neural cells is still *** signaling pathways,such as Hes1 signaling molecular,have an important role in neural stem cell *** this study,the putative involvement of the Hes1 signaling pathway in the neuronal differentiation of BMSCs was investigated through RNA interference(RNAi).We isolated BMSCs from adult rats with complete bone marrow blood *** found that Hes1-siRNA could efficiently block expression of Hes1 in *** induction,immunocytochemistry,reverse transcriptase-PCR and Western blot analysis,all indicated that the expression of neuron-specific markers such as neuron-specific enolase was higher in Hes1-siRNA BMSCs than that in control *** the level of Mash1 expression increased in Hes1-siRNA BMSCs than that in control *** data indicate that Hes1-Mash1 signaling plays a role in the differentiation of BMSCs into neurons in *** or genetic interference with Hes1 signaling may provide a novel method to obtain neurons for therapeutic use.
