Microarray analysis to examine the gene expression changes in the brains of NR2B induced memory enhancement mice

摘      要:正Glutamate N-methyl-D-aspartate(NMDA) receptors play a pivotal role in different forms of *** dysfunction of NMDA receptors contributes to the pathology of CNS *** further investigate the underlying mechanisms of NMDA receptors in the brain processes,we analyzed the gene expression profiles of the hippocampus between NR2B overexpression induced memory enhancement mice(Tg mice) and the *** results revealed that 249 genes were significantly affected in Tg mice,which mainly involved in neurotransmission,signal transduction,cytoskeleton,hormone activity and ***,the intracellular calcium channel proteins, ryanodine receptor(RyR) 1 and 3,as well as functional related proteins such as histidine rich calcium binding protein (Hrc) and triadin 2(Trdn) were ***,our results showed that Homer-lc protein was also increased in Tg mice,and formed a complex with RyR protein in mice brain,suggesting that Homer-lc might be an important modulator on both intracellular calcium signaling and overall neuronal signaling by interacting with NMDA receptor and RyR *** blot and Real-time PCR results showed that the expression of phospho-CREB, c-fos,immediate early genes Egr2 and Egr4 were also up-regulated in Tg *** studies demonstrated that chronic increased activation of NMDA receptors modified the expression of a large number of genes,which may provide important clues for better understanding the molecular mechanism of NMDA receptor modulated learning and memory and CNS disorders.
