Comparison of the cardioprotective effects of purified Danshen and Sanqi mixture with ramipril for the management of acute myocardial infarction in rats

摘 要:Danshen and Sanqi are believed to be able to invigorate blood and transforms blood stasis, thus traditionally, they have been indicated for chest pain, i.e. the commonest symptom of heart attack or acute myocardial infarction (AMI). In this study, the cardioprotective effects of a purified mixture of 70% Danshen (Salvia miltiorrhiza) and 30% Sanqi (Panax notoginseng) (DSSQ) were investigated by comparison with Ramipril (an angiotensin-converting enzyme inhibitor) using rat models of *** Wistar rats were divided into four groups - Sham (10 rats), DSSQ (32 rats), Vehicle (24 rats), and Ramipril (24 rats). Rats from each group were fed orally with 1 ml/kg/day of Water,100mg/kg/day of DSSQ, 1ml/kg/day of Water, and 1mg/kg/day of Ramipril solutions respectively. After one week, the latter three groups were subjected to permanent ligation of left anterior descending coronary artery (LAD) to induce AMI. Sham group was also subjected to the same surgical procedure, except LAD-ligation. Survival rate was determined after 24h of operation. Survived rats were treated with the same regimens for another one week before being sacrificed to collect their heart and liver samples for further *** contrast to Vehicle group with a survival rate of 50.0%, Ramipril and DSSQ groups showed much higher survival rates (66.7% and 62.5%) after AMI. Their differences were statistically significant (p=0.049). Infarct size was smaller for Ramipril (0.302±0.072) and DSSQ (0.315±0.070) groups, compared to Vehicle group (0.336±0.035). Cardiac hypertrophy indicators - left ventricle to whole heart ratio and whole heart to body weight ratio of Ramipril and DSSQ groups were also lower than those of Vehicle group. However, their differences were statistically insignificant (p0.05). Besides, the hepatic anti-oxidant enzymes (CAT, GPx, GST & SOD) and TBARs activities in DSSQ and Ramipril groups were higher than those of Vehicle group.
