Screening, Identification and Function Research on the Candidate Genes of Vaccine against Schistosomiasis

摘 要:Aim Schistosomiasis is a severe world public-health problem, with more than 600 million people exposed to infection and nearly 200 million people infected. At the end of 2003, there were 110 counties from 7 provinces in China involved in the epidemic areas of schistosomiasis with a sharp increase of the patient number and the area of Oncomelania hupensis habitat recently. It’s necessary to find out effective vaccine molecules against this plague. A number of different schistosome antigens have been identified, about 15% of which are strongly protective and deemed promising, but they do not reach the level close to sterile immunity after vaccination with irradiated cercariae. The national schistosome vaccine unitive test carried out in 2000 showed the natural molecular vaccine (Vaccine No.1) constructed by Wang SP et al. was the best vaccine of 16 vaccines, with a protective immunity level close to that of the irradiated cercariae vaccine. Based on the results above-mentioned, we prepared the sera from rabbits vaccinated with irradiated cercariae (VRS) and the sera from pigs vaccinated with the natural molecular vaccine. Schistosoma japonicum (Sj) cercaria cDNA library was screened by the two sera and 5 novel genes were found. Further functional studies on the protective effects of the 2 from these novel genes were performed to value the potentiality of being the effective component of vaccine against schistosomiasis, and to lay a solid foundation for the construction and field-test of the vaccine in the future.
