IL-21 is produced and augments T cells effector functions in emphysema mice

摘 要:Rationale Interleukin(IL)-21 has been reported to be involved in many Th1-associated ***,the alteration and immune regulation of IL-21 in emphysema remains unknown. Objectives To investigate the immunomodulatory effects of IL-21 on T lymphocytes in emphysema. Methods The protein levels of IL-21 was assayed by enzyme-linked immunosorbent assay,the frequencies of CD4IL-21,CD4IL-21R and CD8IL-21R T cells were analyzed by flow cytometry in the peripheral blood,respectively. The immunomodulatory effects of IL-21 on CD4 T cells(or CD8 T cells ) from CS-exposed mice and air-exposed mice were also analyzed. Measurements and Main Results The protein levels of IL-21 in peripheral blood of CS-exposed mice were significantly higher than those of air-exposed *** frequencies of CD4IL-21,CD4IL-21R and CD8 IL- 21R T cells were significantly increased in peripheral blood of CS-exposed mice compared with those in air-exposed ***,the production of IFN-gama,perforin and granzyme B by CD8 T cells and production of IFN-gama by CD4T cells were increased by in vitro stimulation with rIL-21,and these effects were greater in CS-exposed mice than in air-exposed mice. Conclusions Our data suggested that increased IL-21 and IL-21R expression may be involved in the development of emphysema,possibly by augmenting T cells effector functions.
