Zinc finger protein HZF1 promotes the proliferation of K562 cells and inhibits its apoptosis

摘      要:We have previously identified and characterized a novel zinc finger protein gene HZF1 and demonstrated its important role in erythroid and megakaryocytic differentiation of K562 cells (Leukemia,20:1109-1116,2006).We recently examined other functions of *** lentiviral vector-mediated gene transfer we obtained stable cell lines with HZF1 over-expression (K562-GFP-HZF1) and stable control cell lines that integrated GFP only(K562-GFP).The cell cycles of both cell lines were analyzed by detection of intracellular DNA and the result demonstrated that *** promoted the S/G2 phase transition of cell cycle. Furthermore,both cell lines were exposed to the same concentration of AsO and we found that the cells over-expressing HZF1(K562-GFP-HZF1) could more effectively resist apoptosis induced by AsO compared with the control cells(*** accordance with the prospect that HZF1 is analogous to some of anti-apoptosis proteins regulated by mir34c,we identified two potential sites for mir34c targeting within the 3’UTR of HZF1 mRNA *** dual-luciferase reporter experiment and Western-blot assay we further proved that mir34c could knockdown the HZF1 ***,our results showed that zinc finger protein HZF1 could promote cell proliferation by facilitating the S/G2 phase transition of cell cycle and inhibit cell apoptosis induced by AsO.
