摘 要：Background and Aim:Eastern equine encephalitis(EEE) is a significant public health concern as its pathogen EEEV, a mosquito-borne virus, could cause both human and equine encephalitis with high case fatality *** E2 protein is one of the main immunogenic protein of EEEV and plays an important role in the protective immune *** there is no licensed vaccine or treatment for human infection other than supportive therapy so it is essential to develop therapeutics that effectively ameliorate or prevent diseases associated with *** there is no report about the determinate epitope map of EEEV E2 ***, we can develop and study on EEEV E2 protein-specific monoclonal antibodies(m Abs) and further identify the epitopes of the E2 *** and Results:In this study, 51 EEEV E2 protein-specific MAbs were prepared and *** also comprehensively mapped the E2 protein by pepscan with MAbs and antisera(polyclonal antibodies, PAbs) using Western blot and peptide ELISA.18 linear B cell epitopes that are targeted by murine antibody responses were *** them, there were 7 overlapping epitopes(E2-1,11+12,14,24,25,28,33) recognized by both MAbs and PAbs, 9 epitopes(E2-2,11,12,16,18,21,31,32,34) were only recognized by PAbs and 2 epitopes(E2-22,25+26), located at 211-226 aa and 248-259 aa, were only recognized by *** specificity identification of the epitopes corresponding with MAbs showed that there were 7 common epitopes of EEEV and 2 common epitopes of EEEV/*** among them, there were 4 EEEV antigenic complexspecific, EEEV subtype Ⅰ / Ⅱ / Ⅲ / Ⅳ(1-16 aa, 248-259 aa, 271-286 aa, 321-336aa) and 3 EEEV type-specific epitopes(subtype Ⅰ at 108-119 aa, subtype Ⅰ / Ⅱ / Ⅲ at 231-246 aa and subtype Ⅰ / Ⅳ at 211-226aa).The 2 common epitopes of EEEV/VEEV were at 131-146 aa and 241-256 ***:The MAbs of EEEV E2 protein prepared and different types of epitopes identified in our study will establish the foundation for th
