Isolation and Characterization of Maize WRKY Genes Involved in Abiotic Stress Tolerance

摘      要:In this paper,we describe the cloning and characterization of two genes induced by stress factors from maize,and designed MaWRKY7+、MaWRKY80 *** protein with 328 amino acids encoded by MaWRKY79 while MaWRKY80 protein was composed of 458 amino *** domains analysis shows that both of the two genes belong to The WRKY *** localization indicated these two proteins are located in the nucleus of onion cells through transient expression *** addition, we cloned and analyzed the promoter region of MaWRKY79 gene,after we obtain the transgenic lines,GUS testing;the results show that the promoter is stress inducible *** two genes were expressed at low levels in maize seedling leaves and roots, but their expression levels were induced by stress factors,such as PEG,high salinity,and low temperature *** results suggested that they might be involved in abiotic stress *** order to investigate biological functions,overexpression constructs of these two genes were made and transformed into Arabidopsis by Agrobacterium - *** results indicated that the transgenic lines overexpressing MaWRKY79 improved tolerance to drought,low temperature and high ***,AAB1、ICE1、ABA2、ABI1gene was upregulated in the transgenic lines;Transgenic lines overexpressing MaWRKY80 improved tolerance to drought stress and ABI5、SOS 1、ABI1、ABI2、ICE1、DRFB、RD19A gene was upregulated in the transgenic *** results suggested that MaWRKY79 and MaWRKY80 gene play important role in stress *** addition,After Determination of physiological index,we find that praline value were upregulated in both of the transgenic lines In conclusion,these results suggest that MaWRKY79、MaWRKY80 may function in the development and in the signal transduction pathway that involved in abiotic stresses.
