摘      要:Object:To study the neuroprotective effects of Notoginsenoside Rl(NGRl) against HO-induced oxidative stress in PCI2 cells and the underlying mechanism. Results:In this study,for the first time,we clearly showed protective effects of NGRl against HO-induced oxidative stress in PC 12 ***(10 and 100μM) pretreatment obviously inhibited HO-induced ROS accumulation and oxidative stress in *** pretreatment could also suppress HO-induced the imbalance of Bcl-2 family protein,disruption of mitochondrial membrane potential and activation of caspase-3 as well as inactivation of Akt/Nrf2 ***,we fatherly clarified a novel underlying *** treatment alone could up-regulate HO-1 in dose- and time- dependent manners,and NGRl-mediated up-regulation of HO-1 was abolished by transfection with Nrf2 siRNA and supplementation with a PI3K specific inhibitor *** treatment alone could also induce phosphorylation of Akt,facilitate the nuclear accumulation of Nrf2,and enhance the binding activity of Nrf2-ARE in dose- and time- dependent *** importantly, NGRl-mediated activation of Nrf2/ARE patliway was suppressed by transfection with Akt siRNA and ***,NGRl-mediated protection was also attenuated by a HO-1 specific inhibitor Snpp and LY294002. Conclusion:NGRl provides neuroprotective effects against oxidative stress in PC 12 cells by up-regulation of HO-1 through Akt/Nrf2 pathway.
