The role of neuroglobin and cytoglobin against oxidative stress-induced toxicity and hypoxia-induced death in Caenorhabditis elegans models

摘 要:正Objective Neuroglobin(NGB) and cytoglobin(CGB) are newly discovered members of the oxygen-binding globin superfamily,which are expressed in neuronal tissues and many other tissues,*** difference expression patterns between NGB and CGB suggest that the two globins may play different *** investigate whether the these two globins expressed in neurons play distinct neuronal-specific functions,we focused on a simple organism model, ***,to shed light on the protective roles of the globins in *** *** overexpressing human NGB(hNGB) or CGB(hCGB) in the entire nervous system under the control of an unc-119 promoter were *** protective roles of the two globins in neurons were identified by paraquat sensitivity assay and hypoxic sensitivity assay. Results(1) human NGB transgenic worms showed paraquat *** contrast,hCGB transgenic worms showed similar paraquat sensitivity to control worms.(2) Overexpression of human NGB in *** can rescue the paraquat sensitive phenotype of mutation tm2825 strain of *** gene gib-13,which displayed sequence similarity to human neuroglobin.(3) Human NGB/human CGB transgenic worms failed to suppress hypoxia-induced death.(4) The mutation strain(tm2825) displayed hypoxia *** The two globins expressed in neurons play distinct neuronspecific function in *** *** transgenic worms showed paraquat-resistant phenotype but not hypoxia resistance,which is important for us to understand the relationship between reactive oxygen species(ROS) and hypoxia injury.
