摘 要:Touch sensation is essential for behaviors ranging from environmental exploration to social interaction,but the underlying mechanisms are largely *** Drosophila larvae,two types of sensory neurons,class ⅡI and class IV dendritic arborization(da)neurons,tile the body *** mechanotransduction channel Piezo in class IV da neurons is essential for sensing noxious mechanical stimulus but is not involved in gentle *** we report that class ⅡI da neurons are touch sensitive and contribute to gentle touch sensation,based on electrophysiological recording,calcium imaging and behavioral *** further identify Nomp C(no mechanoreceptor potential C),a member of the transient receptor potential(TRP)family of ion channels,as a mechanotransduction channel for gentle *** C is highly expressed in class ⅡI da neurons and required for their ***,ectopic Nomp C expression confers touch sensitivity to the normally touch insensitive class IV da *** only is Nomp C critical for gentle touch to elicit behavioral responses,Nomp C expression in Drosophila S2cells gives rise to mechanosensitive channels whose ion selectivity can be altered by Nomp C mutation,indicating Nomp C is a pore-forming subunit of a mechanotransduction *** study establishes Nomp C as a bona fide mechanotransduction channel that satisfies all four criteria and mediates gentle touch sensation,and suggests that different mechanosensitive channels might be utilized to sense gentle touch versus noxious mechanical stimulus.
