Effect of Conjugated Linoleic Acid on Tumor Cell Growth and Angiogenesis

摘 要:A dietary fatty acid,cis-9,trans- 11-conjugated linoleic acid(CLA) derived from dairy products and meats of ruminant animal, has been shown to be a potent antitumour agent in vitro and in *** is capable of inducing tumour cell apoptosis by up-regulation of p53,p21 and Bax in some human cancer cell ***,its precise mechanism in anti-tumour progression remains elusive. In this study,we found that a high dose of CLA(144μM) but not a low dose(36μM) decreased Akt and eNOS phosphorylation in GP2D(a human colon cancer cell line) after vascular endothelial growth factor(VEGF-50 ng/mL) *** also demonstrated that CLA at the high dose completely inhibits VEGF-induced human umbilical vein endothelial cell(HUVEC) capillary-like tube formation after 7 days co-culture with murine fibroblast *** conclusion,the CLA might play an important role on tumer cell growth and angiogenesis by inhibiting the eNOS and AKT phosphorylation in relation to nitric oxide (NO) ***,we are defining a direct effect of CLA on tumour proliferation, apoptosis and migration as well as its significant biological function in different tumour cell lines.
