Essential role of PKC-delta in radial migration of cortical neurons

摘      要:正The layered structure of the mammalian neocortex is formed through a well-coordinated radial migration of cohorts of post-mitotic neurons during early *** proteins have been identified as essential for the migration of neurons and the formation of stereotyped cortical ***,how the newborn neurons are regulated to migrate or to terminate their migration at their destined layers remains *** we examined the contribution of a member of novel PKC in the migration of newborn neurons to their final layer in developing rat cortex. Using in utero electroporation of small interference RNAs(siRNA),we found that specific down-regulation of PKC-delta severely impaired the targeting of the newborn neurons to their destined layers,without affecting proliferation of neural progenitor cells or the differentiation of newborn *** also found that down-regulation of cdk5 lead to similar migration defect to that of *** antibody against p35 immunoprecipitated ***,brain- neurotrophic factor(BDNF) strongly increased the phosphorylation level of both PKC-delta and *** of TrkB receptor also lead to migration defects of cortical ***,the PKC-delta signaling may interact with p35-cdk5 complex and regulate the proper radial migration of newborn cortical neurons.
