Comparative Evaluation of Nanoemulsion based Transdermal system with Marketed Patch

摘 要:The present work covers the comparative evaluation of nanoemulsion based transdermal patches of Diclofenac Diethylamine (DDEA) with marketed patch. The comparative evaluation was done for the physical parameters like thickness, tackiness, and peel adhesion test and in-vitro release. The in-vitro release characteristics was compared using USP Dissolution apparatus 6 (Cylinder Type) and Keshary-Chein cells using cellulose mixed ester membrane. Dissolution studies were carried out in pH 6.5 phosphate buffer solution at stirring rate of 50rpm at 32 ± *** sampling points selected for the studies were 0, 2, 4, 12 and 24 hours. Nanoemulsions based transdermal therapeutic systems were found to have similar tackiness but better peel adhesion property than marketed patches. It was found that a nanoemulsion based transdermal patches with smaller size and reduced thickness can have a similar release profile to that of marketed patch with a good F2 value. The incorporated drug was released by the Fickian type of diffusion, as is evident by the slope values of less than 0.5 for the plots of log amount released Vs log time. The slope values of marketed and synthesized patches are 0.4 and 0.16 respectively. The correlation values of marketed and synthesized patches are 0.9941 and 0.9945 respectively. From comparative evaluation of physical parameters and in-vitro release profile, we can conclude that the synthesized nanoemulsion based transdermal patches could be a novel smaller sized transdermal dosage form for effective therapy but additional extradermal tests and experiments should be performed before the formulation is used in humans.
