Probucol downregulates heat shock protein 70 expression and decreases vulnerablilty of atrial fibrillation in alloxan-induced diabetic rabbits

摘      要:Objective Probucol is a diphenolic compound with anti-oxidant and anti-inflammatory properties that reduces atherosclerosis and *** benefit of probucol on atrial fibrillation(AF) prevention remains *** sought to investigate the effects on atrial heat shock protein 70(HSP 70) expression and in AF promotion in alloxan-induced diabetic rabbits. Methods Twenty alloxan-induced diabetic rabbits were *** of these were treated with probucol at a dose of 1000 mg/d for 8 weeks and the remaining were left untreated while 10 additional healthy rabbits served as *** Langendorff perfused rabbit hearts were used to evaluate interatrial conduction time(IACT) and vulnerability to *** blotting was performed with left atrial tissues for heat shock protein(HSP70) *** also performed histology examination to measure atrial fibrosis and measured plasma oxidative stress biomarkers. Results Atrial HSP70 expression and AF inducibility were significantly increased in the diabetic rabbits than controls(8/10 s vs 1/10,P0.05).Probucol significantly downregulates atrial HSP 70 expression,reduced AF vulnerablilty in the treated rabbits(3/10 vs 8/10,P0.05).Moreover,Probucol attenuated atrial structural remodeling reducing the IACT and the atrial interstitial fibrosis as well. Probucol increased plasma superoxide dismutase(SOD) activity and on the other hand decreased malondialdehyde(MDA),Medullary oxide enzyme(MPO) and catalase(CAT) levels. Conclusions Probucol downregulates atrial HSP 70 expression and attenuates AF promotion in alloxan-induced diabetic *** seems to relate to its powerful antioxidant activity in this experimental model.
