Immune analysis on mtHSV mediated tumor therapy in HSV-1 seropositiye mice

摘 要:Many patients will be HSV-1 seropositive before treatment with mtHSV for tumor therapy. Balb/c mice were injected intraperitoneally with mtHSV and wtHSV. HSV seropositive and naive Balb/c mice were established sarcoma in the armpit of left forelimb and treated subsequently with mtHSV intratumorally. Tumor mean growth ratios were inhibited greatly in wtHSV seropositive mice, but there was no difference between mtHSV seropositive mice and naive mice. Compared with different doses of mtHSV on tumor therapy, the effect would be better if the dose of mtHSV was higher. The effects between single and multiple treatments with the same dose of mtHSV were not essentially different. We concluded HSV seropositivity could not weaken but enhance the effect on mtHSV-mediated tumor therapy. The cytotoxicity of PBMC from HSV seropositive and naive mice to S-180 cells in vitro showed the killing percentage mice was the highest in wtHSV seropositive. Cytokines transcription level of PBMC showed the IFN-gamma were enhanced remarkably in the early and late infected time in HSV seropositive mice, which is an important factor to inhibit the tumor growth or eliminate the tumor directly, and TNF-alpha also played important role in anti-tumoral function in wtHSV seropositive mice.
