Bioinformatics Analyses Combined with Synthesized Peptides Reveal an Antigenic Determinant of a Membrane Protein of LCDV-cn

摘      要:Lymphocystis disease virus has a broad spectrum of hosts in both freshwater and saltwater fish. The disease causes considerable economic loss to fish fanning industries around China. Type species of the virus named as LCDV-1 isolated from a flounder has its genome been completely sequenced. Based on the genomic information of LCDV-1, two gene/ORFs coding for a major capsid protein and a membrane (myristylated) protein were selected and their counterparts in LCDV-cn were amplified and sequenced. After homology verification, we applied an integrated methodology to screen potential antigenic determinants or mimotopes of the proteins. Six mimotopes generated from the bioinformatics analysis were synthesized and tested in a competitive ELISA. One out of the six peptides, which derived form ORF-29 of LCDV-1, showed strong competence with three specific monoclonal antibodies for LCDV-cn. The results indicate that bioinformatics technology may provide a rapid, economic and efficient way in epitope, mimotope screening and mapping.
