摘 要:Objective Study the biological activity of recombinant porcine interferon alpha1. Methods Protein purity identification,N-terminal amino acid sequencing,titer determination and main physical &chemical properties of the developed recombinant porcine interferonα1 have been carried out according to the recombinant human interferonα1b national standards made by the Ministry of ***,we studied its safety and pharmacokinetic in Landrace pigs. Results The protein purity of developed recombinant porcine interferonα1 identified by SDS-PAGE electrophoresis and HPLC,is more than 95%.N-terminal amino acid sequence,CDLPQTHSLAHTRALR,is also in line with the porcineαinterferon *** titer of developed recombinant porcine interferonα1,which was detected with recombinant humanα1b(rhIFN-α1b) as a reference standard,is more than 1.0×10 IU/*** main physical and chemical properties:the biological activity of recombinant recombinant porcine interferonα1 can be neutralized by porcine interferon amonoclonal antibody(Abcam company);the recombinant recombinant porcine interferonα1 is extremely sensitive to trypsin,while insensitive to acid(pH2.0) and 56℃,for example,its biological activity is preserved in 52.6%when put at 56℃for heat *** porcine clinical safety study results show that,in each group(intravenous,intramuscular and subcutaneous group),after injected with different doses of the recombinant porcine interferonα1 for 2 weeks,porcine appearance and behavioral characteristics were normal,body temperature and weight changes are within the normal range,the vital organs anatomy revealed no obviously lesions,the changes of routine blood test and blood biochemical parameters were normal,no obvious toxic side *** pharmacokinetic model of recombinant porcine interferon alpha1 intramuscular injection group and the subcutaneous injection group are consistent with one-compartment open model and features firstorder eliminate kinetics simultaneously,Tmax were(2
