Effects of vagus nerve stimulation on heroin seeking in a model of relapse in rats and the underlying neurobiological mechanisms

摘 要:正Objective The present study was performed to investigate the effects of vagus nerver stimulation(VNS) on drug-seeking elicited by cues or heroin,and observed the expression of transcriptional factors in NAc after the VNS *** The rats were trained to acquire heroin self-administration under fixed ratio reinforcement schedule (FR1).After daily 4h self-administration training session for 10 consecutive days,rats that acquired heroin self-administration were divided into 3 groups:operation group,low intensity vagus nerver stimulation group and high intensity vagus nerver stimulation *** group was treated correspondingly once a day for 10 days,following treatment,rats were put into the previously training cages with environmental *** the last treatment and extinct training,the rats were tested for reinstatment of the drug seeking induced by conditional cues or heroin in the trainning cages for *** the trial,Western blotting was applied to detect the expression of p-CREB protein in related brain *** Active pokes induced by the conditioned cues in low intensity VNS group(56.9±10.9) and high intensivity VNS group(46.7±7.1) were decreased markedly than the operation group(166.2±20.9)(P0.01) in the two hours of test *** induced by heroin priming,active pokes of L-VNS group(13.0±4.8) and H-VNS group(15.2±5.2) were decresed clearly than the operation group(32.5±4.7)(P0.01).The inactive pokes between three groups are insignificantly different either in conditional cues or heroin induced test session(P0.05).In the Western blotting experiment,p-CREB was incresed dramatically in NAc of the low or high intensity VNS group compared with the operation group either in conditional cues or heroin induced test *** The present data demonstrated that vagus nerver stimulation could inhibit the relapse of the drug seeking behavior induced by conditioned cues and heroin *** mechanism may be involved in the
