Identification of new skin whitening ingredient: Hei Lao Hu extract reduces melanin synthesis

摘 要：We isolated various extracts from Hei Lao Hu, a well known traditional Chinese medicine, and found inhibitory effect on melanogenesis. Among them, an extract dose-dependently inhibited melanin synthesis in melan-a, mouse melanocytes. The extract showed more effectiveness than arbutin;melanin synthesis inhibitory activity treated with 2ppm of the extract was better than that with 50 ppm of arbutin. Using further high performance liquid chromatography(HPLC) analysis, we identified a novel compound, kadsuralignan F with strong melanin synthesis inhibition in melan-a cells(IC50=2.7ppm). Western blot analysis demonstrated that kadsuralignan F significantly reduced melanogenic proteins including tyrosinase, tyrosinase related protein(TRP)-1, and TRP-2. These results suggested Hei Lao Hu has potential as an effective skin whitening agent.
