Interaction between dopaminergic and cholinergic transmission in the nucleus accumben mediates the heroin relapse induced by conditioned cues

摘 要:正The mesolimbic dopamine system projects from VTA to the nucles accumbens(NAc) and is a key neural substrate regulating drug *** previous experiment showed that the enhacement of acetylcholine in NAc blocks the heroin seeking induced by conditioned *** present study was to observe the activation patterns of NAc neurons marked by FosB during the instatement induced by condtioned cues after heroin *** rats were self- administered heroin infusion under escalating FR1-5 reinforcement schedule for two *** reinstatement was tested after the withdrawal for 2 *** active responding induced by conditioned cues which associated previsously with heroin infusion was higher than that indcued by ***,pretreatment with an acetylcholinestase inhibitor could decreased the responding induced by conditioned cues compared the conditioned cues exposed *** FosB positive nuclei was mainly located in the TH positive neurons in the NAc shell,but did not change in the NAc core during conditioned cues induced *** contrast,the relative intensity of ChAT expression in NAc core and shell decreased when exposed to conditioned *** pretreatment with the AChE inhibitor increased the ChAT and FosB expression in NAc core and shell,and also inhibited the increase of TH and FosB positive neurons in NAc shell induced by conditioned *** data demonstrated that heroin withdrawal may result in the neuroadapation of dopaminergic and acetylcholinergic transmission,and acetylcholine play a reciprocal role in the heroin relapse induced by conditioned cues.
