ROS does not contribute to acute development of nitroglycerine tolerance in rat aortas

摘 要:正The development of nitroglycerine tolerance limits its clinical efficacy of nitric oxide(NO)*** cellular mechanisms have been proposed to explain this phenomenon,one of which is an exaggerated production of reactive oxygen specious(ROS).The present study examined whether ROS contribute to the occurrence of nitroglycerine tolerance which is induced acutely in aortas isolated from male Sprague-Dawley *** aortas were exposed to 30μmol/L nitroglycerine(with and without pre-incubation of ROS inhibitors) for 90 min and washed out 4 times during the following 60 min before phenylephrine was added to induce a steady contraction. Subsequently,nitroglycerine was added cumulatively(3 nmol/L-50μmol/L)to evoke ***-induced relaxations were severely impaired after 90-min pre-exposure to *** with apocynin,tempol and tiron plus DETCA
