摘 要:AIM Metabolic variations in serum of senes- cence accelerated mouse (SAM) and the effect of Liuwei Dihuang decoction (LW) on serum metabolites were in- vestigated with metabonomic techniques utilizing [H] NMR spectroscopy and pattern recognition analysis. METHODS Female SAM-resistance/1 (SAMR1) and SAM-prone/8 (SAMP8) at the ages of 12-month-old were employed and orally administered with LW for consecutive 30 *** in control groups were given the equal volume of saline *** metabolites were identified on 500-MHz [H] NMR *** integrated data were normalized to the total integrals of a spectrum and princi- pal components analysis (PCA) was performed using SIM- CA-P+10.04 *** were preprocessed with Pareto scaling prior to *** The results in- dicated that [H] NMR spectra of serum samples from SAMR1 and SAMP8 were obviously different and the vari- ations contributed to the differences were lactate, glucose, choline, inosine,*** LW intervention, the metabo- nomic spectra of sera from SAMP8 were affected and LW improved the metabolic disturbance of SAMP8 in many ways of metabolism by regulating the levels of serum me- *** The differences between ser- um metabolites from SAMR1 and SAMP8 would provide clues for aging and functional disturbance of SAMP8 and the improvements of LW on SAMP8 might owing to the regulation on metabolism of *** metabonomic tech- nique could be used as a potentially powerful tool to inves- tigate the biochemical changes of certain physiopathologic conditions such as aging, and also suitable for studying the pharmacologic action of traditional Chinese prescription.
