Genetic variation of TAP gene and the outcomes of hepatitis C virus infection in drug users

摘 要:Objective Transporter associated with antigen processing(TAP) has essential roles in the antigenpresenting systems,translocating antigenic peptides from the cytosol into the endoplasmic reticulum. Genetic polymorphisms of TAP gene may be involved in the outcomes of HCV infection.A casecontrol study was conducted to test this *** Total 173 drug users(DUs) with HCV persistent infection,189 DUs with HCV self-limiting infection,and 225 DUs with HCV uninfection were obtained during the one-year period(2006.01-2006.12).Antibodies to HCV antigens were detected by enzyme-linked *** presence of viral particles in the serum was determined by reverse -transcriptase polymerase chain reaction(RT-PCR).The genotypes of six polymorphisms of TAP gene were analyzed by ARMS-PCR *** Of the six polymorphisms,one of which(TAP2-379) was observed to have statistically significant association with recovered HCV infection(P0.05),two of which(TAP1-637 and TAP2-379) were observed to have statistically significant association with persistent HCV infection(P0.05),and one of which(TAP1-637) was observed to have statistically significant association with outcomes of HCV infection(P0.05).The extended haplotype constructed with TAP polymorphisms was analyzed among the healthy control,recovered and persistent *** TAP2 haplotype H(Ile-Ala-Arg-Thr) and K(Ile-Thr-Arg-Thr) were shown to present significant difference between the recovered and control groups(P0.05).TAP1 haplotype B(Ile-Gly),D(Val -Gly),and TAP2 haplotype E(Val-Thr-Arg-Thr),H(De-Ala-Arg-Thr) were shown to present significant differences between the persistent and control groups(P0.05).The frequency of TAP1 haplotype B(De-Gly) or D(Val-Gly) was found increased significantly in persistent group,in comparison with that in recovered group(P0.05).Conclusions These findings suggest a potential role of TAP2 gene as a candidate gene for susceptibility to HCV *** the genetic polymorphism
