Preliminary Evaluation of Injectable Silk Fibroin/Polyurethane Composite Hydrogel For Nucleus Pulposus Replacement

摘 要:*** degenerative disc disease,an injectable hydrogel can fill the degenerate area completely, reduce the risk of implant migration and subsequent loss of height of the intervertebral disc,and minimize the surgical ***,we propose a method of preparing an novel injectable and in situ curable silk fibroin/ polyurethane(SF/PU) composite hydrogel by chemical cross-linking under physiological conditions,for initial orthopaedic applications. Methods,the study will investigate(1) synthesis of silk fibroinpolyurethanecomposite hydrogel(2) its unconfined compressive behavior,(3) impact of height on the biomechanical properties,(4) its rheological properties(5) Cell proliferation assay,(6) SEM observation,(7) its implantability,and (8 ) the visibility of the hydrogel on X-rays,CT and magnetic resonance imaging(MRI). *** hydrogel have a gelling time of 5-30 minat room temperature (25°C) and atmospheric *** the 5-30 min ’working time,’ the mixture was injected into moulds using a 14-gauge needle. Under unconfined compression,silk fibroin polyurethane composite hydrogels show a non-linear stress- strain *** tangent modulus value at 15%,20%and 25%strain was 0.30,0.34 and 0.39MPa, respectively. Our preliminary findings on the impactof height indicated that the tangent modulus values increased with increase ofthe cylinder height. The visco-elastic properties ofthe hydrogel showed the lossmodulus G was always smaller thanthestorage modulus G’.The phase shift angle was less than45°*** current hydrogel was stiffer and more elastic than that of natural NP tissue. Cell proliferation assay using bone marrow stromal cells(R-BMSC),showed positive cell viability and increased proliferation over a seven-day period in culture.
