In vitro assessment of chemical genotoxicity by p53-luc/GFP-based reporter system

摘      要:DNA-damaging genotoxic stress could recruit p53 acting as the guardian of the genome and the p53 genotoxic stress response is partly regulated at the transcription level. We constructed human and murine p53 promoter-driven luciferase and green fluorescent protein (GFP) reporter gene expression plasmids to explore possibility of developing a p53-luc/GFP based reporter gene assay system for assessing genotoxicity of chemicals. NIH-3T3 cells transfected with the reporter vectors were treated with anti-cancer drugs and the expression of the reporter gene in the cell lysates was assayed. Luciferase and GFP were greatly induced in NIH 3T3 cells by genotoxic agents indicating the p53-luc/GFP based reporter gene assay system could be applied in the detection and assessment of genotoxicity of environmental toxins.
